NO AND N2O FLUXES FROM A BELGIAN FOREST AFFECTED BY ELEVATED NITROGEN DEPOSITION Gayana Botánica
Boeckx,Pascal; Vervaet,Hilde; Cleemput,Oswald van.
Nitrous oxide (N2O) and nitric oxide (NO) are important atmospheric trace gases, and soils (including forest soils) are a substantial source of both gases. In forests subjected to elevated N deposition these emissions are considered as indirect emissions from agricultural N sources (mainly NH3) or combustion (e.g. NOx from traffic). However, knowledge about N2O and NO exchange between forests and the atmosphere is scarce. The aim of this study was to determine N2O and NO emission rates from a deciduous forest in Belgium, receiving a high N-deposition (ca. 40 kg N ha-1 yr-1). In April 2000, the NO emission from the forested location was 17.3 (± 1.2) and 17.05 (± 0.7) ng NO-N m-2 s-1, respectively, for the soil including the organic layer and the mineral...

Genotoxic evaluation of sodium nitroprusside in Aspergillus nidulans Genet. Mol. Biol.
Chiuchetta,Simone Jurema Ruggeri; Castro-Prado,Marialba Avezum Alves de.
The exogenous nitric oxide donor, sodium nitroprusside, evaluated the recombinogenic potential of nitric oxide. Drug inhibited mycelial growth and conidiation in A757 Aspergillus nidulans master strain. Two heterozygous diploid strains, one wild (uvsH+//uvsH+) and the other defective to DNA repair (uvsH//uvsH) were used for recombinagenesis tests. Sodium nitroprusside recombinogenic effect was evaluated by the induction of homozygosis of recessive genes, originally present in heterozygous condition. Results show that sodium nitroprusside (40 muM, 80 muM and 160 muM) is effective in inducing mitotic crossing-over in diploid cells of A. nidulans.

Comparison of haemocytic parameters among flat oyster Ostrea edulis stocks with different susceptibility to bonamiosis and the Pacific oyster Crassostrea gigas ArchiMer
Comesana, Pilar; Casas, Sandra M.; Cao, Asuncion; Abollo, Elvira; Arzul, Isabelle; Morga, Benjamin; Villalba, Antonio.
Farming of the flat oyster Ostrea edulis in Europe is severely constrained by the protozoan Bonamia ostreae. The introduction of the resistant species Crassostrea gigas has been a relief for the farmers, while the pilot programmes to select O. edulis strains resistant to bonamiosis performed in various countries can be seen as a promising strategy to minimise the effects of bonamiosis. However, the physiological bases of this differential susceptibility remain unknown. A search for an explanation of the intra and interspecific differences in oyster susceptibility to bonamiosis was accomplished by comparing some immune parameters among various O. edulis stocks and C gigas. On December 2003, naive and Bonamia-relatively resistant flat oysters from Ireland....

The impact of insecticide resistance on Culex pipiens immunity ArchiMer
Vezilier, Julien; Nicot, Antoine; De Lorgeril, Julien; Gandon, Sylvain; Rivero, Ana.
Because of their role as vectors of diseases, the evolution of insecticide resistance in mosquitoes has been intensively investigated. Insecticide resistance is associated to a wide range of pleiotropic effects on several key life-history traits of mosquitoes such as longevity and behavior. However, despite its potential implications in pathogen transmission, the effects of insecticide resistance on mosquito immunity have received little, if any, attention. Here, we investigate the impact of insecticide resistance in Culex pipiens, an epidemiologically important vector of a wide array of pathogens. Using both isogenic laboratory strains and field-caught mosquitoes, we investigate the impact of two main insecticide resistance mechanisms (metabolic...

Bohlouli,Somayeh; Khazaei,Mozafar; Rabzia,Arezou; Khazaei,Mohammad Rasool; Sadeghi,Ehsan.
Endometriosis is an estrogen-dependent disease in reproductive age women. Adiponectin and Nitric oxide (NO) have an important role in physiologic functions especially in human reproductive system. Levels of NO increased in the endometriosis patients but serum adiponectin levels decreased in woman with endometriosis. The aim of this study was to determine adiponectin effect on nitric oxide secretion by cultured normal and endometriotic human endometrial stromal cells. In this experimental study, normal (n= 10) and endometriotic endometrial biopsies (n= 10) were taken in sterile condition. Stromal cells isolated and cultured in in DMEM/ F12 medium and treated with adiponectin concentrations (0, 10, 100, and 200 ng/ml) for 24 and 48 hours. NO assay was done...

ENALAPRIL AND VERAPAMIL ATTENUATE THE AORTIC WALL REMODELING IN NITRIC OXIDE DEFICIENCY International Journal of Morphology
Meirelles Pereira,Leila Maria; Gonçalves Bezerra,Daniele; Mandarim-de-Lacerda,Carlos Alberto.
Forty mature male rats (Wistar strain) were maintained alive for 40 days, separated in four groups of five rats each (Control, L-NAME, L-NAME + Enalapril, L-NAME + Verapamil). Three aortic wall parameters were analyzed, as follows: the Q A(SMN) (the number of the tunica media smooth muscle nuclear profiles per area), the S V(lamellae) (surface density of the aortic lamellae), and the aortic wall thickness (AWT). Rats of the L-NAME group showed high blood pressure and cardiac hypertrophy. The AWT, the Q A(SMN) and the S V(lamellae) showed the same tendency among the groups. The increased AWT, or the reduced Q A(SMN) and the S V(lamellae) in l-NAME rats were not completely prevented by the administration of enalapril or verapamil. Hypertension causes the AWT...

ORSI,R.O.; FUNARI,S.R.C.; SOARES,A.M.V.C.; CALVI,S.A.; OLIVEIRA,S.L.; SFORCIN,J.M.; BANKOVA,V..
Propolis has been the subject of several recent studies, with the aim of elucidating its biological and pharmacological properties. Propolis has a well-known antimicrobial activity as well as antioxidant, antitumoral, antiinflammatory, and regenerative properties, but literature about its effects on the immune response is scarce. The goal of this work was to evaluate the propolis effect on macrophage activation by oxygen (H2O2) and nitrogen (NO) metabolite determination. Propolis was produced by africanized honeybees and hydroalcoholic solutions were prepared at different concentrations. Peritoneal macrophages were obtained from male BALB/c mice and culture cells were stimulated in vitro with propolis or interferon-gamma (IFN-gamma). In the in vivo assay,...

Hydrogen sulfide: a new endogenous player in an old mechanism of plant tolerance to high salinity Acta Botanica
da-Silva,Cristiane J.; Modolo,Luzia V..
ABSTRACT High salinity affects plants due to stimulation of osmotic stress. Cell signaling triggered by nitric oxide (NO) and hydrogen sulfide (H2S) activates a cascade of biochemical events that culminate in plant tolerance to abiotic and biotic stresses. For instance, the NO/H2S-stimulated biochemical events that occur in plants during response to high salinity include the control of reactive oxygen species, activation of antioxidant system, accumulation of osmoprotectants in cytosol, induction of K+ uptake and Na+ cell extrusion or its vacuolar compartmentation among others. This review is a compilation of what we have learned in the last 10 years about NO participation during cell signaling in response to high salinity as well as the role of H2S, a new...

Arginine metabolism in uricotelic species - doi: 10.4025/actascianimsci.v32i4.10990 Animal Sciences
Fernandes, Jovanir Inês Müller; Universidade Federal do Paraná; Murakami, Alice Eiko; UEM.
Due to the lack of a complete urea cycle, uricotelic species, such as broilers, are not able to synthesize de novo arginine (Arg), thus depending exclusively on dietary Arg. High levels of dietary lysine (Lys) increase the demand for Arg because of the antagonistic relationship between these amino acids. The Arg-Lys antagonism promotes an expressive increase in the renal Arg activity and consequently induces the degradation of Arg and the decrease in the activity of glycine amidinotransferase, an enzyme that uses Arg in the synthesis of muscle creatin. Arg is considered an important modulator of immunological and physiological processes. The degradation of Arg produces ornithine, a precursor of polyamines that are key to cell division, DNA synthesis, and...

ABSTRACT Zanthoxylum rhoifolium Lam. (Rutaceae) has been traditionally used in the treatment of microbial infections and parasitic diseases. In the present study, the antileishmanial effect induced by the ethanol extract of stem barks from Z. rhoifolium (ZR-EEtOH) and its n-hexane fraction (ZR-FHEX) on infection and infectivity of murine macrophages by promastigote forms of Leishmania amazonensis were investigated. In different set of experiments, macrophages or promastigotes were pretreated with ZR-EEtOH or ZR-FHEX at non-lethal concentrations for 24 hours, and then macrophages were submitted to infection by promastigotes. Moreover, their effects on activation of macrophages, as well as on the DNA content, size and number of promastigotes by flow...

Immunology and immunopathology of African trypanosomiasis Anais da ABC (AABC)
Vincendeau,Philippe; Bouteille,Bernard.
Major modifications of immune system have been observed in African trypanosomiasis. These immune reactions do not lead to protection and are also involved in immunopathology disorders. The major surface component (variable surface glycoprotein,VSG) is associated with escape to immune reactions, cytokine network dysfunctions and autoantibody production. Most of our knowledge result from experimental trypanosomiasis. Innate resistance elements have been characterised. In infected mice, VSG preferentially stimulates a Th 1-cell subset. A response of gd and CD8 T cells to trypanosome antigens was observed in trypanotolerant cattle. An increase in CD5 B cells, responsible for most serum IgM and production of autoantibodies...

ABA regulation of post-germination desiccation tolerance in wheat cultivars contrasting in drought tolerance Anais da ABC (AABC)
KOUR,SATINDER; ZHAWAR,VIKRAMJIT K..
ABSTRACT Post-germination desiccation tolerance (DT) was studied in two wheat cultivars. Effect of pretreatment of abscisic acid (ABA)/ osmotic/ salt/ heat stress was also studied. One day (d)-old seedlings of wheat cultivars PBW 644 (drought tolerant) and PBW 343 (drought susceptible) were exposed to ABA/stress treatment for next 1 d, desiccated for 4 d and subsequently rehydrated for 4 d. Biomass, protein, water, protein carbonyls (oxidative toxicity) and nitric oxide (NO) levels were measured in 2 d (treated), 6 d (desiccated), 10 d (rehydrated) seedlings. Vegetative reactive oxygen species (ROS)/ NO-pathways were studied under normal condition and ABA supply by supplying ROS/NO scavengers. Desiccation caused water loss and increased oxidative toxicity....

Removal of the cortical projections alters expression of NOS in the different cell types of the superficial layers of the superior colliculus in rats Anais da ABC (AABC)
TENÓRIO,FRANK; GIRALDI-GUIMARÃES,ARTHUR; MENDEZ-OTERO,ROSALIA.
Nitric oxide has several biological roles and nitric oxide synthase (NOS) is expressed in the nervous system, and co-localizes with NADPH-diaphorase. The superficial layers of the superior colliculus (SC), which receive retinal and cortical inputs, present NADPH-d staining in a sub-population of neurons that include all cell types. We have previously shown, by NADPH-diaphorase, that eye enucleation alters the intracellular distribution of NOS. Here, we studied the effect of cortical ablation on NOS expression by neurons in collicular superficial layers. Our results show that cortical ablation alters the proportion of different NOS-positive cell types, but not the intracellular distribution of the enzyme.

Effects of rotational culture on morphology, nitric oxide production and cell cycle of endothelial cells Biocell
Chaojun,Tang; Wu,Xue; Linqi,Ye; Xiang,Xie; Guixue,Wang.
Devices for the rotational culture of cells and the study of biological reactions have been widely applied in tissue engineering. However, there are few reports exploring the effects of rotational culture on cell morphology, nitric oxide (NO) production, and cell cycle of the endothelial cells from human umbilical vein on the stent surface. This study focuses on these parameters after the cells are seeded on the stents. Results showed that covering of stents by endothelial cells was improved by rotational culture. NO production decreased within 24 h in both rotational and static culture groups. In addition, rotational culture significantly increased NO production by 37.9% at 36 h and 28.9% at 48 h compared with static culture. Flow cytometry showed that...

Nitric oxide synthase activity in tissues of the blowfly Chrysomya megacephala: Fabricius, 1794 Biocell
Faraldo,A. C.; Sá-Nunes,A; Faccioli,L. H.; Del Bel,E. A.; Lello,E.
Although insects lack the adaptive immune response of the mammalians, they manifest effective innate immune responses, which include both cellular and humoral components. Cellular responses are mediated by hemocytes, and humoral responses include the activation of proteolytic cascades that initiate many events, including NO production. In mammals, nitric oxide synthases (NOSs) are also present in the endothelium, the brain, the adrenal glands, and the platelets. Studies on the distribution of NO-producing systems in invertebrates have revealed functional similarities between NOS in this group and vertebrates. We attempted to localize NOS activity in tissues of naïve (UIL), yeast-injected (YIL), and saline-injected (SIL) larvae of the blowfly Chrysomya...
